You can download iOS 4.3.3 all day long - but you can't install it on your iPad.
iOS 4.3.5 is the most current firmware for the iPad. And, there is no going back to older versions unless you've saved what is known as SHSH blobs. See here for more info on those: SHSH Blobs - Frequently Asked Questions - UPDATED
The odds are that if you're just now asking about 4.3.3 - you don't have the blobs saved...
Overall, biggest difference is that iOS 4.3.5 has some security patches, while 4.3.3 doesn't. Oh, and you can use 4.3.3 to jailbreak the iPad2. Other than that, there's really no difference.
