No. You will be able to update 5.0.1 that way, but iOS 5 will still require you to connect to a computer running iTunes.
Apple stores will doing this as a free service I understand from an employee.

The first release of iOS 5 will be 5.0.0. It will include the ability to update future versions - iOS 5.0.1 for instance - over the air without a computer attached to iTunes.
The current version 4.n does not support over the air updates.

In iOS 5.... You have to install iOS 5 first to get those features...
