In iTunes, go to Edit > Preferences > Devices. Under Devices, you should see the last backup you have. Delete that backup. Then, try to do a backup again. It should work...
Hope that helps. Let us know how you get on.

There are no backups there! I went to the Devices tab but there were no backups listed on the little window over there.
I am afraid I wont be able to update to OS 5 too if the problem continues...
Take a look at this post over at the Apple site: iTunes: "Backup could not be saved on the computer" alert message
You've already tried the 1st step (removing the backup from within iTunes) - which you couldn't do since no backup was there. So, go to the applicable (PC or Mac) address specified in the link and see if there are any backups there. It may be that they are damaged and iTunes just can't see them.
If there are any at the address, copy them (just in case) and then delete them. Then, try running the backup again (right click, "Backup") within iTunes. Hopefully, that'll do it.
Let us know how that works.

Try doing a full reset of your ipad. Hold down the power and home buttons until the apple logo appears.
